Ketepatan prosedur pemeriksaan sangat berpengaruh terhadap hasil pemeriksaan. Aktivitas enzim dipengaruhi oleh beberapa faktor salah satunya yaitu waktu inkubasi. Jika waktu inkubasi kurang dari waktu standar optimumnya, maka tidak akan bereaksi sempurna. Penelitian ini bertujuan untuk mengetahui perbedaan lamanya waktu inkubasi terhadap kadar glukosa darah metode enzimatik yang diinkubasi pada suhu ruang selama 5 menit, 10 menit, 30 menit, 60 menit dan 90 menit. Penelitian ini dilakukan di Laboratorium Hematologi Kimia Klinik Jurusan Analis Kesehatan Politeknik Kesehatan Kemenkes Banjarmasin bulan Desember 2018. Jenis penelitian yang digunakan adalah Quasi Eksperiment dengan rancangan Posttest Only with Control Group Design. Sampel penelitian ini menggunakan serum dari 5 responden yang memiliki kadar glukosa darah normal. Rata-rata hasil penelitian lamanya waktu inkubasi selama 5 menit 78,244 mg/dl; 10 menit 83,870 mg/dl; 30 menit 84,436 mg/dl; 60 menit 84,516 mg/dl; 90 menit 84,65 mg/dl. Hasil penelitian ini menunjukkan semakin lama waktu inkubasi semakin tinggi kadar glukosa darah. Berdasarkan hasil uji Repeated Anova menunjukkan ada perbedaan bermakna antara kadar glukosa darah yang diinkubasi selama 5 menit dengan kontrol 10 menit yaitu 0,001 < α (0,05). Pada waktu inkubasi 30 menit, 60 menit, dan 90 menit menunjukkan tidak ada perbedaan terhadap kontrol 10 menit dengan nilai signifikansi > α (0,05). Bagi peneliti selanjutnya dapat melakukan penelitian mengenai perbedaan lamanya waktu inkubasi terhadap kadar glukosa darah metode enzimatik menggunakan sampel serum yang abnormal.

The accuracy of the examination procedure is very influential on the results of the examination. The activity of the enzyme is influenced by several factors, one of which is incubation time. If the incubation time is less than the optimum standard time, it will not react perfectly. This research is aimed to determine the differences in the length of incubation time with enzymatic methods of blood glucose levels, which were incubated at room temperature for 5 minutes, 10 minutes, 30 minutes, 60 minutes and 90 minutes. This research is usually done in hematology laboratory – chemist clinic on Analis Kesehatan Poltekkes Kemenkes Banjarmasin in December 2018. The type of research used was QuasiExperiment with the design of Posttest Only with Control Group Design. The sample of this research used serum from 5 respondents who had normal blood glucose levels. The result of research the incubation time were 5 minutes 78,244 mg/dl; 10 minutes 83,870 mg/dl; 30 minutes 84,436 mg/dl; 60 minutes 84,516 mg/dl; 90 minutes 84.65 mg/dl. The result of the research showed that the longer the incubation time the higher the blood glucose level. Based on the result of analysis test Repeated ANOVA, there was a significant difference between the blood glucose levels incubated for 5 minutes with 10 minutes control, which was 0.001 < α (0.05). At the incubation time of 30 minutes, 60 minutes, and 90 minutes showed no difference in the control of 10 minutes with a significance value > α (0.05). For the next researcher will be able to do the research on the differences in length of incubation time with blood glucose levels in the enzymatic method using abnormal serum samples.
