Pemeriksaan laboratorium klinik merupakan penunjang diagnosis. Salah satu tahapan dalam pemeriksaan laboratorium diantaranya tahap pra analitik yang memberikan kontribusi kesalahan dengan frekuensi 77,1 % (Goswami, 2010). Pemeriksaan faal hemostatis yaitu Activated Partial Thromboplastin Time (APTT) menggunakan plasma sitrat harus dilakukan dalam waktu 4 jam (Woodhams 2001). Penundaan pemeriksaan dapat menyebabkan perubahan faktor pembekuan darah terutama faktor V dan VIII yang menurun (Sacher 2012). Tujuan penelitian ini untuk mengetahui ada tidaknya pengaruh lama penyimpanan plasma sitrat terhadap nilai APTT. Jenis penelitian yaitu quasi eksperiment dengan rancangan postest only control group design. Hasil rata-rata pemeriksaan APTT dengan bahan pemeriksaan yang didiamkan pada suhu ruang selama 0 jam (segera), ditunda 2 jam, 4 jam, 6 jam, dan 8 jam berturut-turut yaitu 33,5 detik, 34,2 detik, 34,9 detik, 37,0 detik, dan 39,5 detik. Berdasarkan uji statistik regresi linier didapatkan nilai signifikasi = 0,000 yang membuktikan ada pengaruh lama penyimpanan sampel plasma sitrat terhadap penetapan APTT yaitu 45,1 %. Disarankan bagi petugas laboratorium agar mengetahui stabilitas sampel plasma.

ABSTRACT Effect Long Storage of Plasma Citrate at Room Temperature Against Determination of Activated Partial Thromboplastin Time (APTT) Using the Automatic Method

Clinical laboratory examination is a diagnostic support. One of the stages in a laboratory examination is the pre-analytic stage which contributes an error of 77.1% (Goswami, 2010). Hemostatic physiology examination, namely Activated Partial Thromboplastin Time (APTT) using plasma citrate, must be done within 4 hours (Woodhams 2001). Delayed examination can cause changes in blood clotting factors, especially factors V and VIII are decreased (Sacher 2012). The purpose of this study was to determine whether there is an influence of plasma citrate storage duration on APTT values. The type of research is quasi experiment with posttest only control group design. The results of the average APTT examination with samples that were left to stand at room temperature for 0 hours (immediately), delayed 2 hours, 4 hours, 6 hours, and 8 hours respectively were 33.5 seconds, 34.2 seconds, 34.9 seconds, 37.0 seconds, and 39.5 seconds. Based on the linear regression statistical test obtained a significance value = 0,000 which proves that there is an influence of the duration of storage of plasma citrate samples on the determination of APTT that is 45.1%. It is recommended for laboratory personnel to know the stability of plasma samples.
