PENGARUH LAMA PENGERINGAN PREPARAT APUS DARAH TEPI PADA SUHU 37°C TERHADAP MORFOLOGI SEL DARAH MERAH (Erythrocyte)

Sediaan apus darah tepi merupakan pemeriksaan untuk menghitung jenis morfologi sel darah merah meliputi bentuk dan warna. Salah satu faktor penentu dalam hal ini yaitu teknik pembuatan sediaan apus darah serta faktor-faktor lainnya seperti waktu, pewarnaan dan suhu. Pengeringan menggunakan suhu tinggi dapat menyebabkan sel darah merah menjadi rusak. Penelitian ini bertujuan untuk mengetahui pengaruh lama pengeringan preparat apus darah tepi pada suhu 37°C terhadap morfologi sel darah merah. Jenis penelitian ini adalah Quasi Eksperiment dengan Rancangan penelitian Posttest Only Control Group Design. Spesimen menggunakan darah yang berjumlah 6 orang berjenis kelamin perempuan dengan kriteria yang memiliki hemoglobin normal dan tidak ada kelainan morfologi eritrosit. Hasil penelitian didapatkan persentase rata-rata kenaikan perubahan morfologi eritrosit yang mengalami kerusakan dilihat dari bentuk ekinosit pada sediaan apus darah tepi pada pengeringan suhu 37°C selama 3 menit = 0,64%; 20 menit = 3,85%; 40 menit = 18,64%; 60 menit = 47,42%. Sedangkan yang dilihat dari bentuk akantosit didapatkan pengeringan selama 3 menit = 1,21%; 20 menit = 3,09%; 40 menit = 11,63%; 60 menit = 26,21%. Adapun yang dilihat dari warna hiperkrom pada pengeringan suhu 37°C selama 3 menit = 1,11%; 20 menit = 1,54%; 40 menit = 3,89%; 60 menit = 5,73%. Sedangkan yang dilihat dari hipokrom pada pengeringan suhu 37°C selama 3 menit = 1,39%; 20 menit = 2,07%; 40 menit = 4,53%; 60 menit = 7,42%. Berdasarkan hasil analisis data menggunakan uji Regresi Linear diperoleh nilai signifikansi 0,000 yang berarti P < 0,05 maka disimpulkan bahwa ada pengaruh lama pengeringan preparat apus darah tepi pada suhu 37°C terhadap morfologi eritrosit dilihat dari ekinosit, akantosit, hiperkrom dan hipokrom. Berdasarkan hasil penelitian tersebut disarankan untuk melakukan penelitian lebih lanjut tentang pengaruh lama pengeringan preparat apus darah tepi terhadap ukuran morfologi eritrosit.

EFFECT OF LONG DRYING PREPARATE OF PERIPHERAL BLOOD SMEAR AT THE 37 ° C TEMPERATURE ON THE MORPHOLOGY OF RED BLOOD CELL (Erythrocyte)

The peripheral blood smear is an examination to calculate the type of of red blood cells morphology including shape and color. One of the determining factors in this case is the technique of making blood smears and other factors such as time, staining and temperature. Drying using high temperatures can damage the red blood cells. This study aims to determine the effect of drying duration of peripheral blood smear at 37 ° C on the morphology of red blood cells. This type of research is a Quasi Experiment with Posttest Only Control Group Design research design. The specimens used are blood from 6 people of female sex with criteria that have normal hemoglobin and no morphological abnormalities of erythrocytes. The results showed an average percentage increase in morphological changes in erythrocytes that suffered damage seen from the form of echinocytes in the preparation of peripheral blood smears at a drying temperature of 37 ° C for 3 minutes = 0.64%; 20 minutes = 3.85%; 40 minutes = 18.64%; 60 minutes = 47.42%. Whereas seen from the form acanthocyte obtained drying for 3 minutes = 1.21%; 20 minutes = 3.09%; 40 minutes = 11.63%; 60 minutes = 26.21%. As for what is seen from the color of hyperchrome at a drying temperature of 37 ° C for 3 minutes = 1.11%; 20 minutes = 1.54%; 40 minutes = 3.89%; 60 minutes = 5.73%. While those seen from hypochrome at drying temperature of 37 ° C for 3 minutes = 1.39%; 20 minutes = 2.07%; 40 minutes = 4.53%; 60 minutes = 7.42%. Based on the results of data analysis using the Linear Regression test obtained a significance value of 0,000, which means P <0.05, it was concluded that there was an influence of drying duration of peripheral blood smear at 37°C on the morphology of erythrocytes seen from echinocytes, acanthocytes, hyperchromes and hypochromes. Based on these results it is recommended to conduct further research on the effect of drying time of peripheral blood smear preparations on the morphological size of erythrocytes.
