Fiksasi adalah tahapan pertama dalam histoteknik pada pengolahan jaringan.
Fiksasi adalah suatu proses menjaga jaringan dari degenerasi, dekomposisi,
pembusukan, distorasi dan autolisis jaringan setelah dikeluarkan dari tubuh dan
harus dilakukan sesegera mungkin. Jenis larutan yang dapat digunakan untuk
fiksasi selain NBF 10% yaitu larutan Alkohol 70%. Alkohol 70% adalah salah satu
larutan fiksatif yang memiliki daya penetrasi yang cepat, murah dan mudah
diperoleh sedangkan NBF 10% mempunyai pH yang netral namun memiliki daya
penetrasi yang lebih lambat. Tujuan penelitian ini untuk mengetahui adanya
perbedaan kualitas pewarnaan Hematoksilin-Eosin pada organ hepar tikus putih
(Rattus norvegicus) yang difiksasi menggunakan larutan Alkohol 70% selama 12
jam, 16 jam dan 20 jam dengan larutan NBF 10% 24 jam sebagai kontrol. Jenis
penelitian True Experiment dengan rancangan Posttest only control group design.
Hasil pewarnaan Hematoksilin-Eosin pada organ hepar tikus putih yang difiksasi
dengan NBF 10% 24 jam (kontrol) diperoleh hasil 100% baik, fiksasi Alkohol 70%
variasi waktu 12 jam diperoleh hasil 83% baik, variasi waktu 16 jam diperoleh hasil
66% baik serta variasi waktu 20 jam diperoleh 56% baik. Disimpulkan bahwa
kualitas sediaan histologi yang difiksasi menggunakan Alkohol 70% variasi waktu
16 dan 20 jam dengan NBF 10% 24 jam ada perbedaan bermakna, sedangkan
sediaan histologi yang difiksasi dengan Alkohol 70% variasi waktu 12 jam dengan
NBF 10% 24 jam tidak ada perbedaan bermakna. Untuk peneliti selanjutnya
disarankan menggunakan larutan yang berbeda atau lama waktu fiksasi yang lebih
bervariasi.

“Comparison of the Quality of White Rat (Rattus norvegicus) Liver Histology
Preparations Based on Variations in Fixation Time Using 70% Alcohol”

Fixation is the first stage in histotechnics in tissue processing. Fixation is a process
of protecting tissue from degeneration, decomposition, decay, distortion and
autolysis of tissue after it is removed from the body and must be done as soon as
possible. The type of solution that can be used for fixation other than 10% NBF is
70% alcohol solution. 70% alcohol is a fixative solution that has fast penetration, is
cheap and easy to obtain, while 10% NBF has a neutral pH but has slower
penetration. The aim of this study was to determine differences in the quality of
Hematoxylin-Eosin staining in the livers of white rats (Rattus norvegicus) which
were fixed using 70% alcohol solution for 12 hours, 16 hours and 20 hours with
10% NBF solution for 24 hours as a control. This type of research is True
Experiment with a Posttest only control group design. The results of Hematoxylin-
Eosin staining on the liver of white mice fixed with 10% NBF for 24 hours (control)
obtained 100% good results, 70% alcohol fixation, a 12 hour time variation
obtained 83% good results, a 16 hour time variation obtained 66% good results. and
a time variation of 20 hours obtained 56% good. It was concluded that there was a
significant difference in the quality of the histology preparations fixed using 70%
alcohol for 16 and 20 hours with NBF 10% 24 hours, while the histology
preparations fixed using 70% alcohol for 12 hours with NBF 10% 24 hours had no
significant difference. For future researchers, research using different solutions or
a more varied length of fixation time can be recommended.
