PENAMBAHAN PROPILEN GLIKOL TERHADAP
STABILITAS KADAR KOLESTEROL”

Pemantapan Mutu Internal laboratorium dilakukan untuk menjamin ketelitian dan
ketepatan hasil pemeriksaan dengan menggunakan bahan kontrol. Kontrol
komersial sangat mahal harganya, sehingga bahan kontrol yang dibuat sendiri dari
kumpulan serum (pooled sera) dapat menjadi alternatif karena lebih hemat biaya.
Pooled sera harus stabil selama penyimpanan, untuk itu diperlukan penambahan
pengawet propilen glikol. Parameter yang digunakan dalam penelitian ini adalah
pemeriksaan kolesterol karena merupakan pemeriksaan rutin yang sering dilakukan
di laboratorium. Tujuan penelitian ini untuk mengetahui pengaruh lama
penyimpanan pooled sera dengan penambahan propilen glikol terhadap stabilitas
kadar kolesterol. Penelitian ini bersifat eksperiment dengan desain pretest -posttest.
Bahan penelitian berupa kumpulan serum dengan kadar kolesterol normal yang
ditambahkan propilen glikol, diukur berdasarkan 6 perlakuan dan 4 kali
pengulangan. Hasil rata-rata kadar kolesterol pooled sera sebelum disimpan (0 hari)
= 146,5 mg/dL dan setelah disimpan 6 hari = 146,0 mg/dL, 12 hari = 145,3 mg/dL,
18 hari = 143,8 mg/dL, 24 hari = 141,5 mg/dL dan 30 hari = 138,8 mg/dL. Uji
Repeated ANOVA menunjukkan adanya perbedaan signifikan kadar kolesterol pada
penyimpanan 24 hari (Sig. 0,009 < 0,05) dan 30 hari (Sig. 0,004 < 0,05) yang
berarti sudah tidak stabil. Demikian disimpulkan bahwa terdapat pengaruh lama
penyimpanan pooled sera dengan penambahan propilen glikol terhadap stabilitas
kadar kolesterol dan pooled sera hanya dapat digunakan sebagai bahan kontrol
pemeriksaan kolesterol selama 18 hari. Saran penelitian selanjutnya, dapat mencoba
pemeriksaan profil lipid lainnya menggunakan kumpulan serum normal atau
abnormal.

Internal Quality Assurance of the laboratory is carried out to ensure the accuracy
and precision of the examination results by using control materials. Commercial
controls are very expensive, so control materials made from pooled sera can be an
alternative because they are more cost-effective. Pooled sera must be stable during
storage, for which the addition of propylene glycol preservative is required. The
parameter used in this study is cholesterol examination because it is a routine
examination that is often done in the laboratory. The purpose of this study was to
determine the effect of pooled sera storage duration with the addition of propylene
glycol on the stability of cholesterol levels. This research is experimental with
pretest-posttest design. The research material was pooled sera with normal
cholesterol levels added with propylene glycol, measured based on 6 treatments and
4 repetitions. The average cholesterol level of pooled sera before storage (0 days)
= 146.5 mg/dL and after storage 6 days = 146.0 mg/dL, 12 days = 145.3 mg/dL,
18 days = 143.8 mg/dL, 24 days = 141.5 mg/dL and 30 days = 138.8 mg/dL.

days storage (Sig. 0.009 < 0.05) and 30 days (Sig. 0.004 < 0.05) which means it is
unstable. Thus it is concluded that there is an effect of the length of storage of
pooled sera with the addition of propylene glycol on the stability of cholesterol
levels and pooled sera can only be used as a control material for cholesterol
examination for 18 days. Suggestions for further research, can try other lipid profile
examinations using normal or abnormal serum pools
