Pemeriksaan di laboratorium klinik harus melakukan pemantapan mutu
internal dengan memeriksa bahan kontrol untuk menjamin ketepatan dan
keakuratan hasil pemeriksaan. Salah satu pemeriksaan yang digunakan adalah
parameter kadar glukosa. Bahan kontrol dapat dibuat sendiri dengan harga yang
relatif murah dibandingkan serum komersial menggunakan serum yang
dikumpulkan atau disebut pooled sera. Pooled sera harus stabil selama periode
penyimpanan, sehingga diperlukan penambahan pengawet natrium azida. Tujuan
penelitian untuk mengetahui pengaruh lama penyimpanan pooled sera dengan
penambahan natrium azida terhadap stabilitas kadar glukosa. Jenis penelitian
adalah eksperiment dengan rancangan pretest-posttest design. Bahan penelitian
berupa kumpulan serum dengan kadar glukosa normal yang ditambahkan natrium
azida sebanyak 24 sampel. Hasil kadar glukosa pada 0 hari = 72, 8 mg/dL, 6 hari
= 71,8 mg/dL, 12 hari = 71,5 mg/dL, 18 hari = 70,8 mg/dL , 24 hari = 67,3
mg/dL, dan 30 hari = 64,5 mg/dL. Uji Repeated ANOVA menunjukkan ada
pengaruh lama penyimpanan pooled sera dengan penambahan natrium azida
terhadap stabilitas kadar glukosa dengan Sig. < α (0,000 < 0,050). Kesimpulan
terdapat pengaruh lama penyimpanan pooled sera dengan penambahan natrium
azida terhadap stabilitas kadar glukosa yang disimpan selama 30 hari sehingga
pooled sera hanya dapat digunakan sebagai bahan kontrol pemeriksaan
laboratorium hingga 18 hari. Penelitian selanjutnya disarankan menggunakan
parameter dan pengawet yang berbeda dari kumpulan serum normal atau
abnormal.

ABSTRACT
The Effect of Storage Duration of Pooled Sera with Addition of
Sodium Azide on the Stability of Glucose Levels

Examinations at clinical laboratories must perform internal quality
assurance by testing control materials to ensure the accuracy and precision of
examination results. One of the tests used is the glucose level parameter. Control
materials can be made in-house at a relatively lower cost compared to
commercial serum by using collected serum, referred to as pooled sera. Pooled
sera must be stable during the storage period, necessitating the addition of the
preservative sodium azide. The aim of this study is to determine the effect of the
storage duration of pooled sera with the addition of sodium azide on the stability
of glucose levels. This research is experimental with a pretest-posttest design. The
research material consisted of collected serum with normal glucose levels to
which sodium azide was added, totaling 24 samples. The glucose levels were: on
day 0 = 72.8 mg/dL, day 6 = 71.8 mg/dL, day 12 = 71.5 mg/dL, day 18 = 70.8
mg/dL, day 24 = 67.3 mg/dL, and day 30 = 64.5 mg/dL. Repeated ANOVA tests
showed an effect of storage duration of pooled sera with the addition of sodium
azide on the stability of glucose levels, with Sig. < α (0.000 < 0.050). The
conclusion is that the storage duration of pooled sera with the addition of sodium
azide affects the stability of glucose levels stored for 30 days, thus pooled sera
can only be used as laboratory control material for up to 18 days. Future
research is recommended to use different parameters and preservatives from
normal or abnormal pooled sera.
