Sediaan apusan darah tipis untuk menentukan morfologi sel eritrosit, leukosit, dan
trombosit dengan pewarnaan Romanowsky pengencer buffer fosfat pH 6,8 yang
dianjurkan penggunaannya namun memiliki harga relatif mahal. Buah naga pengencer
air mineral digunakan sebagai alternatif alami karena buah naga lebih ramah
lingkungan. Tujuan penelitian ini untuk mengetahui perbedaan kualitas pewarnaan
sediaan apusan darah tipis menggunakan zat warna buah naga pengencer air mineral
bermerek dan zat warna Romanowsky pengencer buffer fosfat pH 6,8. Jenis penelitian
adalah observasional laboratorium dengan rancangan comparative study. Berdasarkan
uji Mann-Whitney tidak ada perbedaan kualitas antara zat warna Romanowsky
pengencer buffer fosfat pH 6,8 (A) dan zat warna buah naga pengencer buffer fosfat pH
6,8 (B) signifikansi p=1,000. Berdasarkan uji Kruskal Wallis tidak ada perbedaan
kualitas pewarnaan zat warna Romanowsky pengencer buffer fosfat pH 6,8 (A) dengan
zat warna buah naga air mineral bermerek 1 (C), 2 (D), 3 (E), 4 (F) signifikansi p=1,000
dan begitu juga untuk zat warna buah naga (B) dengan zat warna buah naga air mineral
bermerek 1(C), 2(D), 3(E), 4(F) signifikansi p=1,000 menunjukkan tidak ada perbedaan
kualitas pewarnaan. Persentase kualitas pewarnaan zat warna Romanowsky pengencer
buffer fosfat pH 6,8 didapatkan hasil baik sebesar 100%, zat warna buah naga pengencer
buffer fosfat pH 6,8 didapatkan hasil baik 100%, dan zat warna buah naga pengencer
air mineral 1(C), 2(D), 3(E), 4(F) didapatkan hasil baik sebesar 100%, 97%, 100%,
97%. Disimpulkan bahwa zat warna buah naga pengencer air mineral bermerek dapat
digunakan sebagai alternatif pewarna Romanowsky dan pengencer buffer fosfat pH 6,8.

Thin blood smear preparation to determine the morphology of erythrocyte,
leukocyte, and platelet cells with Romanowsky stain diluent phosphate buffer pH
6.8 which is recommended for use but has a relatively expensive price. Dragon fruit
diluting mineral water is used as a natural alternative because dragon fruit is more
environmentally friendly. The purpose of this study was to determine the difference
in the quality of staining thin blood smear preparations using dragon fruit dye,
branded mineral water thinner, and Romanowsky dye, phosphate buffer diluent pH
6.8. This type of research is observational laboratory with comparative study
design. Based on the Mann-Whitney test, there was no difference in quality between
Romanowsky dye, phosphate buffer diluent pH 6.8 (A) and dragon fruit dye
phosphate buffer diluent pH 6.8 (B), significance p=1.000. Based on the Kruskal
Wallis test, there is no difference in the coloring quality of Romanowsky dye,
phosphate buffer diluent pH 6.8 (A) with dragon fruit dye, branded mineral water
1 (C), 2 (D), 3 (E), 4 (F), significance p = 1,000 and likewise for dragon fruit dye
(B) with dragon fruit dye, branded mineral water 1 (C), 2(D), 3(E), 4(F)
significance p=1,000 indicates no difference in coloring quality. The percentage of
coloring quality of Romanowsky dye diluent phosphate buffer pH 6.8 obtained
good results of 100%, dragon fruit dye diluent phosphate buffer pH 6.8 obtained
good results of 100%, and dragon fruit dye mineral water diluent 1(C), 2(D), 3(E),
4(F) obtained good results of 100%, 97%, 100%, 97%. It was concluded that dragon
fruit dye, a branded mineral water diluent, can be used as an alternative to
Romanowsky dye and phosphate buffer diluent pH 6.8.
