Pemeriksaan Plasma Prothrombin Time (PPT)

Plasma prothrombin time (PPT) merupakan pemeriksaan faal hemostasis untuk
mengetahui kelainan faktor pembekuan darah jalur ekstrinsik. Salah satu faktor yang
dapat mempengaruhi hasil pemeriksaan PPT ialah suhu dan waktu penyimpanan
sampel. Menurut Clinical and Laboratory Standards Institute sampel pemeriksaan PPT
harus diperiksa dalam waktu 24 jam jika disimpan pada suhu 25oC. Penyimpanan
sampel suhu dingin (4oC) menyebabkan teraktivasinya faktor VII sehingga
mempengaruhi nilai PPT. Tujuan penelitian ini untuk mengetahui ada tidaknya
pengaruh suhu dan lama penyimpanan plasma sitrat terhadap hasil pemeriksaan PPT
yang disimpan selama 0 jam, 12 jam, 18 jam, 24 jam serta 30 jam pada suhu 25oC dan
suhu 4oC. Jenis penelitian ini yaitu quasi eksperiment dengan rancangan postest only
control group design. Hasil rata-rata pemeriksaan PPT dengan lama penyimpanan
plasma sitrat selama 0 jam, 12 jam, 18 jam, 24 jam serta 30 jam pada suhu 25oC
berturut-turut yaitu 14,2 detik, 15,1 detik, 15,9 detik, 17,7 detik dan 21,3 detik
sedangkan pada penyimpanan suhu 4oC nilai PPT berturut-turut yaitu 14,2 detik, 13,8
detik, 13,3 detik, 12,8 detik, dan 12,1 detik. Berdasarkan hasil uji regresi linear
didapatkan nilai signifikansi sebesar 0,000 (p < α) yang berarti terdapat pengaruh suhu
dan lama penyimpanan plasma sitrat terhadap hasil pemeriksaan PPT dengan besar
pengaruh 82,9% pada penyimpanan suhu 25oC dan 71,9% pada penyimpanan suhu
4oC. Diharapkan bagi petugas laboratorium untuk memperhatikan stabilitas sampel
khususnya pemeriksaan PPT.

Effect of Temperature and Citrate Plasma Storage Time to the Plasma
Prothrombin Time (PPT) Results

Plasma prothrombin time (PPT) is a hemostasis examination to determine
extrinsic blood clotting abnormalities. One of the factors that can affect the results of
the PPT examination is the temperature and storage time of the sample. According to
the Clinical and Laboratory Standards Institute the PPT examination sample must fit
within 24 hours if stored at 25oC. Very cold sample storage (4oC) causes activation of
factor VII which affects the PPT value. The purpose of this study was to determine
whether there was an effect of temperature and storage time of citrate plasma on the
results of PPT examination which were stored for 0 hours, 12 hours, 18 hours, 24 hours
and 30 hours at 25oC and 4oC. This type of research is a quasi-experimental design
with a post-test only control group design. The average results of PPT examination
with citrate plasma storage time for 0 hours, 12 hours, 18 hours, 24 hours and 30 hours
at 25oC were 14,2 seconds, 15,1 seconds, 15,9 seconds, 17,7 seconds and 21,3 seconds
while at 4oC temperature storage the PPT values were 14,2 seconds, 13,8 seconds, 13,3
seconds, 12,8 seconds, and 12,1 seconds, respectively. Based on the results of the linear
regression test, it was obtained a significance value of 0.000 (p <α), which means that
there was an effect of temperature and storage time of citrate plasma on the results of
PPT examination with a large effect of 82.9% on storage at 25oC and 71.9% at storage
at 4oC. It is expected that laboratory staff will pay attention to the stability of the PPT
examination sample.
