Perbandingan Produk Enzim Urikase pada Cryptococcus laurentii dan Trichosporon asahii dengan Variasi Suhu dan Lama Inkubasi

Urikase merupakan enzim yang berperan katalisis spesifik untuk oksidasi asam urat. Penggunaan urikase murni memiliki kelemahan, yaitu enzim tidak stabil dan biayanya yang mahal. Urikase dari mikroorganisme menjadi alternatif pengganti urikase murni. Tujuan dari penelitian ini adalah membandingkan produk enzim urikase yang dihasilkan oleh Cryptococcus laurentii dan Trichosporon asahii pada kondisi optimum yaitu suhu 27°C dan lama inkubasi 96 jam. Jenis penelitian ini adalah kuasi eksperimen dengan rancangan Factorial Design. Variabel terikat adalah kadar enzim urikase dan variabel bebas adalah suhu inkubasi 27°C dan 37°Cdengan lama inkubasi 48 jam, 72 jam dan 96 jam. Hasil penelitian menunjukkan Cryptococcus laurentiidan Trichosporon asahii  dapat menghasilkan urikase ditandai dengan munculnya zona bening disekitar koloni pada media padat yang tambahkan asam urat. Diameter zona bening sekitar 0,5-1,5 cm. Diperoleh hasil kadar enzim urikase Cryptococcus laurenti 4,4132 U/ml dan Trichosporon asahii 4,3661 U/ml pada suhu 27°C dengan lama inkubasi 96 jam. Kesimpulan dari penelitian adalah tidak ada perbedaan bermakna (p = 0,245 > α) antara enzim urikase yang dihasilkan oleh Cryptococcus laurenti dan Trichosporon asahii pada kondisi optimum suhu 27°C dengan lama inkubasi 96 jam. Saran kepada peneliti selanjutnya sebaiknya waktu inkubasi diperpanjang sehingga dapat diketahui kapan terjadinya penurunan produksi enzim urikase oleh Cryptococcus laurenti dan Trichosporon asahii.

 ABSTRACT 
 
Comparison of Uricase Enzyme Products in Cryptococcus laurentii and Trichosporon asahii with the Variation of Temperature and Incubation Period

Uricase is an enzyme that plays a role in specific catalysis for uric acid oxidation. The use of pure uricase has disadvantages, which are unstable and their expensive costs. Uricase from microorganisms is an alternative substitute for pure uricases. The aim of this study was to compare uricase enzyme products produced by Cryptococcus laurentii and Trichosporon asahii in optimum conditions at 27°C with 96 hours. This type of research is an experiment with Factorial Design. The dependent variable is the uricase enzyme level and the independent variable is the incubation temperature of 27°C and 37°C with incubation period of 48 hours, 72 hours and 96 hours. The results showed that Cryptococcus laurentii and Trichosporon asahii can produce uricase indicated by the appearance of clear zones around the colonies on solid medium that contain uric acid. The diameter of clear zone is around 0,5-1,5 cm. The results of the levels of uricase enzymes Cryptococcus laurentii 4.4132 U/ml and Trichosporon asahii 4.3661 U/ml at 27°C with 96 hours incubation. The conclusion of the study was no significant difference (p = 0.245> α) between uricase enzymes produced by Cryptococcus laurentii and Trichosporon asahii in optimum conditions at 27°C with 96 hours incubation. Suggestions to the next researcher it would be better if the incubation period be extended so that it can be known when the production of uricase enzymes decreases by Cryptococcus laurentii and Trichosporon asahii.
