PENYIMPANAN

Perhitungan jumlah leukosit metode manual memerlukan reagen larutan 
Turk. Komposisi larutan Turk standar adalah asam asetat glasial 2%, gentian violet 
1% dan aquadest. Jeruk lemon (Citrus limon) memiliki kandungan asam sitrat. 
Asam Sitrat pada jeruk lemon merupakan asam lemah yang mana sama dengan 
Asam asetat. Penelitian ini bertujuan untuk mengetahui perbedaan jumlah leukosit 
dengan menggunakan larutan Turk dan larutan Turk yang dimodifikasi dengan 
perasan air lemon yang segera diperiksa, disimpan 1 minggu, 2 minggu, dan 3 
minggu dengan konsentrasi 80%. Jenis penelitian yang digunakan adalah
experiment dengan rancangan posttest only control group design. Analisis data 
menggunakan uji T-test Dependent, rata-rata jumlah leukosit larutan Turk segera, 
disimpan 1 minggu, 2 minggu, dan 3 minggu yaitu 6.713 sel/mm3
, 6.238 sel/mm3

6.175 sel/mm3
,dan 5.975 sel/mm3
. Rata-rata jumlah leukosit larutan Turk 
modifikasi perasan air lemon segera, disimpan 1 minggu, 2 minggu, dan 3 minggu 
yaitu 6.613 sel/mm3
, 6.125 sel/mm3
, 5.850 sel/mm3
,dan 5.625 sel/mm3
. Pada uji 
statistik larutan Turk dan larutan Turk modifikasi perasan air lemon segera 
didapatkan hasil 0,116 dan pada 1 minggu didapatkan hasil 0,078 maka 
menunjukan tidak terdapat perbedaan. Terdapat perbedaan antara larutan Turk dan 
larutan Turk modifikasi perasan air lemon disimpan 2 minggu dan 3 minggu karena 
nilai signifikansi <0,05. Kesimpulan penelitian ini tidak terdapat perbedaan jumlah 
leukosit pada larutan Turk dan larutan Turk yang dimodifikasi perasan air lemon 
segera diperiksa dan disimpan 1 minggu. Saran penelitian ini untuk peneliti 
selanjutnya dapat melakukan penelitian dengan jenis jeruk atau buah yang berbeda 
sebagai pengganti asam asetat dengan variasi waktu penyimpanan.

DIFFERENCE IN LEUKOSIT AMOUNT USING TURK SOLUTION AND 
TURK SOLUTION MODIFIED WITH LEMON (Citrus limon) WATER 
PRESSURE ON VARIATION OF STORAGE TIME

Manual method leukocyte count requires Turk's solution reagent. The 
composition of the standard Turk's solution is 2% glacial acetic acid, 1% gentian 
violet and distilled water. Lemon (Citrus limon) has citric acid content. Citric acid 
in lemon is a weak acid which is the same as acetic acid. This study aims to 
determine the difference in leukocyte counts using Turk's solution and modified 
Turk's solution with lemon juice that is immediately examined, stored for 1 week, 
2 weeks, and 3 weeks with a concentration of 80%. The type of research used is 
experiment with posttest only control group design. Data analysis using Dependent 
T-test, the average leukocyte count of Turk solution immediately, stored 1 week, 2 
weeks, and 3 weeks are 6,713 cells/mm3, 6,238 cells/mm3, 6,175 cells/mm3, and 
5,975 cells/mm3. The average leukocyte counts of the modified Turk solution of 
lemon juice immediately, stored for 1 week, 2 weeks, and 3 weeks were 6,613 
cells/mm3, 6,125 cells/mm3, 5,850 cells/mm3, and 5,625 cells/mm3. In the
statistical test of Turk's solution and Turk's solution modified lemon juice 
immediately obtained the result of 0.116 and at 1 week obtained the result of 0.078, 
it shows there is no difference. There is a difference between Turk's solution and 
modified Turk's solution of lemon juice stored 2 weeks and 3 weeks because the 
significance value <0.05. The conclusion of this study is that there is no difference 
in the number of leukocytes in the Turk solution and the modified Turk solution of 
lemon juice immediately examined and stored for 1 week. The suggestion of this 
study for further researchers can conduct research with different types of citrus or 
fruit as a substitute for acetic acid with variations in storage time.
