Pemeriksaan hitung jumlah leukosit metode manual memerlukan reagen
Larutan Turk. Larutan Turk terdiri dari larutan asam asetat glasial 2% dan gentian
violet 1%. Pada laboratorium klinik sederhana, ketersediaan reagen Turk untuk
menghitung jumlah leukosit seringkali tidak tersedia atau tersedia tetapi
kadaluarsa. Selain itu Gentian Violet 0,24-5 mg/L dalam 48 jam memiliki sifat
sangat toksik. Penelitian ini bertujuan untuk mengetahui perbandingan hitung
jumlah leukosit menggunakan larutan Turk dengan larutan modifikasi Turk
perasan jeruk nipis (Citrus aurantifolia) dan ekstrak ubi jalar ungu (Ipomoea
batatas L). Penelitian ini bersifat true experiment dengan rancangan posttest only
control group design. Sampel penelitian menggunakan darah kontrol normal.
Rata-rata hasil pemeriksaan hitung jumlah leukosit menggunakan larutan Turk
adalah 7.390 sel/mm3. Hasil rerata hitung jumlah leukosit menggunakan
modifikasi pada jeruk nipis 2% dan gentian violet 1% yaitu 7.080 sel/mm3, jeruk
nipis 3% dan gentian violet 1% yaitu 6.870 sel/mm3, ubi jalar ungu 1% dan asam
asetat glasial yaitu 6.810 sel/mm3, jeruk nipis 2% dan ubi jalar ungu 1% yaitu
7.100 sel/mm3, serta jeruk nipis 3% dan ubi jalar ungu 1% yaitu 6.840 sel/mm3.
Melalui uji statistik Reapeted ANOVA didapatkan nilai sig 0,646 (sig>α).
Kesimpulan: tidak terdapat perbedaan bermakna antara Turk dengan Modifikasi
Turk. Saran: Penelitian ini diharapkan dapat menambah pengembangan ilmu
dibidang Hematologi serta acuan dalam pemanfaatan bahan alami sebagai
pengganti Turk pada pemeriksaan Hitung Jumlah Leukosit.

Manual leukocyte count examination requires Turk's solution reagent.
Turk's solution consists of acetic acid glasial 2% and gentian violet 1% solution.
In simple clinical laboratories, the availability of Turk's reagent for leukocyte
counts is often unavailable or available but expired. In addition, Gentian Violet
0.24-5 mg/L in 48 hours is highly toxic. This study aims to determine the
comparison of leukocyte counts using Turk's solution with a modified Turk's
solution of lime juice (Citrus aurantifolia) and purple sweet potato extract
(Ipomoea batatas L). This research is a true experiment with posttest only control
group design. The research sample used normal control blood. The average result
of the leukocyte count examination using Turk's solution was 7,390 cells/mm3.
The average result of leukocyte count using modification in lime 2% and gentian
violet 1% is 7,080 cells/mm3, lime 3% and gentian violet 1% is 6,870 cells/mm3,
purple sweet potato 1% and acetic acid glasial is 6,810 cells/mm3, lime 2% and
purple sweet potato 1% is 7,100 cells/mm3, and lime 3% and purple sweet potato
1% is 6,840 cells/mm3. Through Reapeted ANOVA statistical test, the sig value
was 0.646 (sig>α). Conclusion: there is no significant difference between Turk
and Modified Turk. Suggestion: This research is expected to add to the
development of knowledge in the field of Hematology as well as a template in the
use of natural substances as a substitute for Turk in the Leukocyte Count
examination.
