Penggunaan bahan kontrol dalam laboratorium medis adalah kunci untuk
memastikan hasil tes yang konsisten dari waktu ke waktu. Namun, biaya tinggi
serum kontrol komersial seringkali menjadi hambatan bagi laboratorium kecil. Oleh
karena itu, penggunaan pooled sera sebagai bahan kontrol alternatif dapat
digunakan. Stabilitas pooled sera harus dijaga dan penggunan propilen glikol dapat
digunakan sebagai pengawet alternatif yang terjangkau. Penelitian ini bertujuan
untuk mengetahui pengaruh lama penyimpanan pooled sera dengan penambahan
propilen glikol 15% terhadap stabilitas kadar SGPT selama 30 hari pada suhu 4°C.

Jenis penelitian ini menggunakan penelitian eksperimen dengan rancangan pretest-
postetst. Variabel penelitian ini adalah perlakuan lama penyimpanan pooled sera

dengan penambahan propilen glikol 15% dan kadar SGPT. Sampel yang digunakan
adalah serum kumpulan yang ditambahkan propilen glikol 15% dari total Pooled
sera (10 ml). Hasil penelitian dengan uji Paired T-Test berdasarkan lama
penyimpanan 0 hari, 6 hari, 12 hari, 18 hari, 24 hari dan 30 hari mempunyai nilai
signifikansi 0,182; 0,058; 0,092; 0,092 dan 0,080 atau P > α (0,05) sehingga
disimpulkan bahwa kadar SGPT pada pooled sera dengan penambahan propilen
glikol 15% dapat stabil hingga hari ke-30. Untuk penelitian selanjutnya, disarankan
melakukan pemeriksaan parameter lainnya menggunakan pooled sera dengan
penambahan propilen glikol 15%, perbandingan antara penggunaan propilen glikol
dengan zat pengawet lainnya, perbandingan penyimpanan pooled sera dengan
penambahan propilen glikol di freezer dan refrigerator, membuat pooled sera dari
serum patologis dan pooled sera dengan penambahan propilen glikol 15% dapat
digunakan sebagai bahan kontrol selain di bidang kimia klinik saja.

The use of control materials in a medical laboratory is key to ensuring consistent test
results over time. However, the high cost of commercial control sera is often an
obstacle for small laboratories. Therefore, the use of pooled sera as an alternative
control material can be used. The stability of pooled sera must be maintained and the
use of propylene glycol can be used as an affordable alternative preservative. This
study aims to determine the effect of pooled sera storage duration with the addition of
15% propylene glycol on the stability of SGPT levels for 30 days at 4°C. This type of
research uses experimental research with pretest-postetst design. The variables of this
study were the treatment of pooled sera storage duration with the addition of 15%
propylene glycol and SGPT levels. The sample used was pooled serum added with 15%
propylene glycol from the total pooled sera (10 ml). The results of the study with Paired
T-Test test based on the length of storage of 0 days, 6 days, 12 days, 18 days, 24 days
and 30 days had a significance value of 0.182; 0.058; 0.092; 0.092 and 0.080 or P>
α (0.05) so it was concluded that SGPT levels in pooled sera with the addition of 15%
propylene glycol can be stable until day 30. For further research, it is recommended
to examine other parameters using pooled sera with the addition of 15% propylene
glycol, comparison between the use of propylene glycol with other preservatives,
comparison of pooled sera storage with the addition of propylene glycol in the freezer
and refrigerator, making pooled sera from pathological serum and pooled sera with
the addition of 15% propylene glycol can be used as a control material other than in
the field of clinical chemistry only.
