Gambaran Kualitas Reagen Malachite Green Dan Methylene Blue Pada 
Pemeriksaan Telur Cacing Dalam Feses Metode Kato-Katz

Kecacingan merupakan salah satu penyakit di Indonesia yang sampai saat 
ini masih sangat tinggi. Metode pemeriksaan kecacingan salah satunya adalah 
metode kato-katz, reagen warna yang digunakan dalam pemeriksaan mikroskopis 
metode Kato-Katz yaitu malacite green dan methylene blue yang dilarutkan 
dengan aquades dan glycerol. Berfungsi sebagai larutan kato atau reagen, dengan
perendaman selama 24 jam. Tujuan penelitian ini adalah mengetahui kualitas 
penyerapan warna selotif dan mengetahui kualitas warna latar belakang 
mikroskopis dengan reagen malachite green dan methylene blue. Jenis penelitian 
ini menggunakan Quasy experiment, dengan rancangan One group design,
Penelitian ini menggunakan variasi waktu perendaman 1, 6, 12, 18 dan 24 jam.
Sampel feses positif didapatkan dari Laboratorium Kesehatan Provinsi 
Kalimantan Selatan. Berdasarkan hasil pemeriksaan diketahui kualitas penyerapan 
warna malachite green pada selotif dengan variasi waktu 1, 6, 12, 18 dan 24 jam 
didapatkan warna light sea green, sedangkan kualitas penyerapan warna 
methylene blue pada selotif dengan variasi waktu 1, 6, 12, 18 dan 24 jam 
didapatkan warna cornflower blue. Hasil kualitas warna latar belakang 
pemeriksaan di mikroskop dengan reagen malachite green pada variasi waktu 1, 
6, 12, 18 dan 24 jam didapatkan warna light sea green, sedangkan reagen
methylene blue pada variasi waktu 1, 6, 12, 18 dan 24 jam didapatkan warna light 
cyan. Kesimpulan penelitian ini kualitas penyerapan warna dan kualitas warna 
latar belakang adalah reagen malachite green yang menghasilkan warna lebih baik
dibandingkan reagen methylene blue. Bagi peneliti selanjutnya disarankan agar 
menambah variabel penelitian seperti kualitas pewarnaan reagen malachite green 
dan methylene blue pada warna telur cacing di mikroskop.

ABSTRACT
Description of the Quality Malachite Green and Methylene Blue Reagents in 
the Examination of Worm Eggs in Feces Kato-Katz Method

Worms is one of the diseases in Indonesia which is still very high. One of 
the worms examination methods is the kato-katz method, the color reagents used 
in the microscopic examination of the Kato-Katz method are malacite green and 
methylene blue dissolved in aquades and glycerol. Serves as a cathode solution or 
reagent, with immersion for 24 hours. The purpose of this study was to determine 
the quality of the color absorption of the adhesive tape and to determine the 
quality of the microscopic background color with malachite green and methylene 
blue reagents. This type of research uses a quasy experiment, with a One group 
design. This study uses a variation of immersion time of 1, 6, 12, 18 and 24 hours. 
Positive stool samples were obtained from the Health Laboratory of South 
Kalimantan Province. Based on the results of the examination, it is known that the 
quality of absorption of malachite green color on tape with time variations of 1, 6, 
12, 18 and 24 hours obtained light sea green color, while the quality of absorption 
of methylene blue color on tape with time variations of 1, 6, 12, 18 and 24 the 
clock got cornflower blue color. The results of the quality of the background color 
examination in a microscope with malachite green reagent at time variations of 1, 
6, 12, 18 and 24 hours obtained light sea green color, while methylene blue 
reagent at time variations of 1, 6, 12, 18 and 24 hours obtained color light cyan. 
The conclusion of this study is that the quality of color absorption and the quality 
of the background color is malachite green reagent which produces better color 
than methylene blue reagent. For further researchers, it is recommended to add 
research variables such as the quality of the staining of malachite green and 
methylene blue reagents on the color of worm eggs in the microscope.
