Menggunakan Buffer Fosfat pH 6,8 dan Air Minum Dalam Kemasan

Metode pewarnaan Sediaan Apusan Darah Tipis (SADT) yang dianjurkan 
oleh The Internasional Council For Standardization in Haematology (ICSH) 
salah satunya adalah Pewarnaan Romanowsky. Larutan pengencer yang digunakan 
dalam pewarnaan Romanowsky adalah buffer fosfat. Buffer sebagai larutan 
pengencer Romanowsky harus diperhatikan ketepatan Power of Hydrogen (pH) 
untuk hasil pewarnaan yang baik. Larutan buffer fosfat pH 6,8 sulit didapat untuk 
rumah sakit atau puskesmas yang jauh dipelosok dan harganya cukup mahal, 
Sehingga sebagai alternatif air minum dalam kemasan bisa digunakan sebagai 
larutan pengencer yang lebih murah dan mudah untuk diperoleh. Bertujuan untuk 
mengetahui perbedaan morfologi sel eritrosit dan sel leukosit Metode 
Romanowsky menggunakan Buffer Fosfat pH 6,8 dan Air Minum Dalam 
Kemasan. Jenis penelitian observasional laboratorium dengan rancangan 
comparative study dengan 6 jenis air minum dalam kemasan dan 3 level darah 
kontrol yaitu high, normal, dan low. Dengan 5 kali pengulangan dalam satu merek 
air minum dalam kemasan. Hasil penelitian morfologi sel eritrosit dan sel leukosit 
pada darah kontrol high, normal dan low menggunakan buffer fosfat pH 6,8 dan 
air minum dalam kemasan merek B, C, D dan F memiliki hasil pewarnaan yang 
baik sedangakan air minum dalam kemasan E dan G memiliki hasil pewarnaan 
yang tidak baik. Berdasarkan uji Wilcoxon morfologi sel eritrosit dan sel leukosit 
didapatkan nilai sig >0,05 untuk air minum dalam kemasan B, C, D dan F
terhadap buffer fosfat (A) menunjukkan tidak ada perbedaan bermakna. 
Disimpulkan bahwa air minum dalam kemasan dapat digunakan sebagai 
pengencer dalam pewarnaan sediaan apusan darah tipis.

ABSTRACT
Differences in the Morphology of Erythrocyte and Leukocyte Cells 
Romanowsky Method Using Phosphate Buffer pH 6.8 and Bottled Drinking 
Water

One of the staining methods recommended by The International Council for 
Standardization in Haematology (ICSH) is the Romanowsky stain. The diluent 
used in Romanowsky's stain is phosphate buffer. Buffer as a Romanowsky diluent 
must be considered for the accuracy of Power of Hydrogen (pH) for good staining 
results. Phosphate buffer solution pH 6.8 is difficult to obtain for hospitals or 
health centers that are far away and the price is quite expensive, so as an 
alternative to bottled drinking water can be used as a diluent solution which is 
cheaper and easier to obtain. Aims to determine the morphological differences 
between erythrocytes and leukocytes. Romanowsky's method uses Phosphate 
Buffer pH 6.8 and Bottled Drinking Water. This type of research is an 
observational laboratory with a comparative study design with 6 types of bottled 
drinking water and 3 control blood levels, namely high, normal, and low. With 5 
repetitions in one brand of bottled drinking water. The results of the research on 
morphology of erythrocyte cells and leukocyte cells in high, normal and low
control blood using phosphate buffer pH 6.8 and drinking water in brands B, C, D 
and F good staining results, while drinking water in packages E and G had good 
staining results. poor staining. Based on the Wilcoxon test, the morphology of 
erythrocyte and leukocyte cells obtained sig values > 0.05 for drinking water in 
packages B, C, D and F against phosphate buffer (A) which showed no significant 
difference. It was concluded that bottled drinking water can be used as a diluent in 
staining thin blood smears.
