ABSTRAK PENGARUH LAMA PENUNDAAN PENGECATAN SETELAH FIKSASI PADA SUHU 37°C TERHADAP BENTUK ERITROSIT Penulis : Septi Putri Nursari Pembimbing: Jujuk Anton Cahyono, Yayuk Kustiningsih Sediaan apus darah tepi merupakan pemeriksaan untuk menghitung jenis morfologi sel, diantaranya eritrosit yang meliputi bentuk dan warna. Salah satu faktor penentu kualitas sediaan apus darah tepi yaitu waktu pengecatan. Penundaan pengecatan setelah fiksasi pada suhu yang tidak stabil dapat menyebabkan eritrosit menjadi rusak. Penelitian ini bertujuan untuk mengetahui pengaruh lama penundaan pengecatan setelah fiksasi preparat apus darah tepi pada suhu 37°C terhadap bentuk sel darah merah. Jenis penelitian ini adalah Quasi Eksperiment dengan rancangan penelitian Posttest Only Control Group Design. Spesimen menggunakan darah yang berjumlah 6 orang berjenis kelamin perempuan dengan kriteria tidak ada kelainan morfologi eritrosit pada analisa sediaan apus darah tepi. Hasil penelitian didapatkan persentase rata-rata kenaikan perubahan bentuk eritrosit yang mengalami kerusakan dengan lama waktu penundaan pengecatan setelah fiksasi 3 menit, 20 menit, 40 menit dan 60 menit pada suhu 37°C dilihat dari bentuk normal sebesar 94,3%, 86,5%, 64,67%, dan 39,5%, bentuk ekinosit sebesar 3,5%, 9,5%, 31,17%, dan 52,3% serta pada bentuk akantosit sebesar 1,25%, 2%, 2%, dan 2,50%. Berdasarkan hasil analisis data menggunakan uji Regresi Linear diperoleh nilai signifikansi bentuk normal dan ekinosit sebesar 0,000, sedangkan nilai signifikansi bentuk akantosit sebesar 0,065. Dapat disimpulkan bahwa ada pengaruh bermakna lama penundaan pengecatan setelah fiksasi pada suhu 37°C terhadap bentuk ekinosit dan normal, sedangkan pada bentuk akantosit tidak ada pengaruh bermakna. Berdasarkan hasil penelitian tersebut disarankan untuk melakukan penelitian lebih lanjut tentang pengaruh lama penundaan pengecatan setelah fiksasi apus darah tepi pada suhu ruang, suhu lembab, dan suhu tinggi terhadap bentuk eritrosit. One of the determinants of the quality of the peripheral blood smear is the delay time. Delaying staining after fixation at an unstable temperature can cause erythrocytes to be damaged. This study aims to determine the effect of the delay in staining after fixation of peripheral blood smears at 37°C on the shape of red blood cells. This type of research is a Quasi experimental research design with Posttest Only Control Group Design. Specimens using blood, amounting to 6 women, with the criteria of no erythrocyte morphological abnormalities in the analysis of peripheral blood smears. The results showed that the average percentage increase in changes in the shape of damaged erythrocytes with a long delay in painting after fixation was 3 minutes, 20 minutes, 40 minutes and 60 minutes at 37°C as seen from the echinocyte shape of 3.5%, 9.5 %, 31.17%, and 52.3%, in the form of acanthocytes 1.25%, 2%, 2%, and 2.50%. Based on the results of data analysis using the Linear Regression test, the significance value of normal and echinocytes was 0.000, while the significance value of acanthocytes was 0.065. It can be concluded that there is an effect of the delay in painting after fixation at 37°C on the normal and echinocytes, while there is no effect on the shape of the acanthocytes. Based on the results of this study, it is recommended to conduct further research on the effect of delayed staining after fixation of peripheral blood smear at room temperature, humid temperature, and high temperature on the shape of erythrocytes.
