Tangan adalah bagian dari tubuh manusia yang biasanya menjadi media penularan 
berbagai mikroorganisme patogen seperti bakteri, virus, dan jamur. Transmisi 
penyakit melalui tangan dapat diminimalisasi dengan menjaga kebersihan tangan, 
misalnya selalu menggunakan hand sanitizer. Lengkuas merah (Alpinia purpurata
(Vieill.) K. Schum) mengandung sifat antibakteri sehingga dapat mengurangi
pemakaian alkohol pada hand sanitizer. Tujuan penelitian ini adalah untuk 
mengetahui efektivitas ekstrak rimpang lengkuas merah sebagai hand sanitizer
terhadap penurunan angka kuman. Jenis penelitian yang digunakan adalah true 
experiment dengan rancangan Pretest-Posttest Control Design, yang terdiri atas 7
perlakuan sebelum dan setelah menggunakan hand sanitizer ekstrak rimpang 
lengkuas merah formula 1, formula 2, formula 3, formula 4, dan formula 5 beserta 
kontrol positif (formula X) dan kontrol negatif (air mengalir) dengan 3 kali 
pengulangan. Hasil penelitian menunjukkan hand sanitizer ekstrak rimpang 
lengkuas merah yang mampu menurunkan angka kuman hanya dari formula 2 dan 
formula 3. Formula 2 menurunkan angka kuman sebesar 3,26×102 CFU/mL 
menjadi 1,5×101CFU/mL, sedangkan setelah pemberian formula 3 angka kuman 
sebesar 2,18×105CFU/mL turun menjadi 7,87×103CFU/mL. Dari hasil penelitian 
ini disimpulkan hand sanitizer ekstrak rimpang lengkuas merah efektif menurunkan 
angka kuman, dengan efektivitas tertinggi sebesar 96,4% pada formula 3. Untuk 
penelitian selanjutnya diharapkan untuk melakukan pengujian secara in vitro
terlebih dahulu pada bakteri mikroflora normal dan melihat apakah ada aktivitas 
antibakteri pada hand sanitizer ekstrak rimpang lengkuas merah.

Effectiveness of Red Galangal Rhizome Extract (Alpinia purpurata (Vieill.) K.
Schum.) as a Hand Sanitizer on Reducing Germ Numbers

Hands are part of the human body which are usually a medium for transmission of 
various pathogenic microorganisms such as bacteria, viruses, and fungi. 
Transmission of disease through hands can be minimized by maintaining hand 
hygiene, for example always using hand sanitizer. Red galangal (Alpinia purpurata
(Vieill.) K. Schum) contains antibacterial properties so that it can reduces the use 
of alcohol in hand sanitizers. The purpose of this study was to determine the 
effectiveness of red galangal rhizome extract as a hand sanitizer to reduce germ 
numbers. The type of research used is a true experiment with a Pretest-Posttest 
Control Design, which consists of 7 treatments before and after using hand 
sanitizers with red galangal rhizome extract of formula 1, formula 2, formula 3, 
formula 4, and formula 5 along with positive control (formula X) and negative 
control (water) with 3 repetitions. The results showed that the red galangal rhizome 
extract hand sanitizer was able to reduce the number of germs only from formula 2 
and formula 3. Formula 2 reduced the germ number by 3.26×102 CFU/mL to 
1.5×101CFU/mL, while after giving formula 3 the germ numbers was 2.18×105
CFU/mL decreased to 7.87×103 CFU/mL. From the results of this study, it was
concluded that the hand sanitizer of red galangal rhizome extract was effective in 
reducing the number of germs, with the highest effectiveness of 96.4% in formula 
3. For further research, it is expected to conduct in vitro tests first on normal 
microflora bacteria and see if there are antibacterial activities in hand sanitizer red 
galangal rhizome extract.
