Buah Naga (Hylocereus Polyrhizus) Sebagai Bahan Alternatif Alami Pewarna 
Wright-Stain pada Sediaan Apusan Darah Tepi

Pemeriksaan Hematologi dalam sediaan apusan darah tepi adalah bertujuan 
untuk mengamati morfologi sel eritrosit, leukosit dan trombosit. Pemeriksaan ini 
sangat dibutuhkan di Laboratorium menggunakan pewarnaan yang dianjurkan oleh The 
International Council For Standardization in Haematology dengan metode WrightStain. Larutan zat warna Wright-Stain termasuk pewarnaan sintetis yang sulit terurai, 
harganya mahal dan tidak ramah lingkungan. Salah satu alternatif alami yaitu 
menggunakan buah naga (Hylocereus polyrhizus) sebagai pengganti larutan WrightStain karena memiliki antosianin yang tinggi, lebih murah dan ramah lingkungan. 
Tujuan penelitian ini yaitu buah naga sebagai bahan alternatif alami pewarna WrightStain pada sediaan apusan darah tepi. Jenis penelitian adalah observasional 
laboratorium dengan rancangan studi deskriptif. Jumlah sampel yang diteliti adalah 2 
yaitu ekstrak buah naga dan kontrol Wright-Stain dan darah kontrol low. Dengan 16 
kali pengulangan dalam setiap 1 perlakuan. Berdasarkan hasil penelitian menggunakan 
zat warna Wright-Stain pada darah kontrol low didapatkan hasil morfologi sel eritrosit 
yang baik, sel leukosit dan trombosit yang baik pada pengamatan mikroskopis. 
Kemudian dengan menggunakan pewarnaan zat warna buah naga didapatkan hasil 
morfologi sel eritrosit yang baik, sel leukosit yang baik dan trombosit yang baik dalam 
pewarnaannya. Sedangkan pada leukosit jenis eosinofil hasil morfologinya tidak baik. 
Disimpulkan bahwa buah naga (Hylocereus polyrhizus) dapat digunakan sebagai 
alternatif alami pewarnaan Wright-Stain dalam sediaan apusan darah tepi.

ABSTRACT
Dragon Fruit (Hylocereus Polyrhizus) as an Alternative Natural Stain to WrightStain in Peripheral Blood Smear

Hematological examination of the peripheral blood smear is an examination 
that aims to observe the morphology of erythrocyte, leukocyte, and platelet cells. The 
examination is considered vital in the laboratory using the stain approved by the 
International Council for Standardization in Hematology with the Wright-Stain 
method. The Wright-Stain solution is categorized as a synthetic stain as it is difficult 
to decompose, expensive, and harmful to the environment. Concerning the negative 
qualities of the synthetic stain, an alternative natural stain up for consideration is to use 
dragon fruit (Hylocereus polyrhizus) as a substitute for Wright-Stain because it is high 
in anthocyanins, cheaper, and environmentally friendly. This study aims to conduct an 
observation using dragon fruit (Hylocereus polyrhizus) as an alternative natural stain 
to Wright-Stain in peripheral blood smear. The type of research employed is laboratory 
observation with a descriptive research design. The number of samples studied was 
two, namely dragon fruit extract and Wright-Stain control and low blood control, done 
with 16 repetitions in each treatment. According to the study results using Wright-Stain 
on low blood control, erythrocyte, leukocyte, and platelet cells were found good on 
observation microscopic. Subsequently, using dragon fruit stain, the results obtained 
also include good morphologies of erythrocyte, leukocyte, and platelet cells in their 
staining. On the other hand, the morphological result of eosinophil leukocytes was not 
good. The study shows that dragon fruit (Hylocereus polyrhizus) can be used as an 
alternative natural stain for Wright-Stain staining in peripheral blood smear 
preparations.
