Pengaruh Variasi Volume Pencucian Terhadap Pembacaan Nilai Absorbansi 
Anti-HbsAg Metode ELISA

Hepatitis B adalah penyakit yang menimbulkan peradangan pada organ hati 
dan biasanya disebabkan oleh Virus Hepatitis B (HBV). HbsAg merupakan ciri 
khas dari infeksi sedangkan Anti-HbsAg merupakan antibodi terhadap HbsAg yang 
muncul ketika penderita pernah terinfeksi HBV pada waktu yang cukup lama atau 
pernah divaksinasi Hepatitis B, metode yang sering digunakan untuk pemeriksaan 
Anti-HbsAg adalah ELISA (Enzyme-Linked Immunosorbent Assay). Penelitian ini 
bertujuan untuk mengetahui pengaruh pada variasi volume pencucian terhadap 
pembacaan nilai absorbansi Anti-HBsAg metode ELISA reader. Jenis penelitian 
ini bersifat eksperimen dengan rancangan Posttest Only with Control Design. 
sampel diambil dengan teknik purposive sampling pada petugas kesehatan 
Puskesmas Martapura Timur yang telah divaksinasi hepatitis B sebanyak 3 orang, 
dilakukan pemeriksaan Anti-HbsAg berdasarkan variasi volume pencucian 
menggunakan metode ELISA reader. Analisis data menggunakan uji Repeated
ANOVA. Hasil penelitian menunjukkan pada variasi volume pencucian yang 
dilakukan yaitu 100 µl, 150 µl, 200 µl, 250 µl, dan 300 µl pada nilai statistik adalah 
0,224 (p>0,05) membuktikan tidak ada pengaruh yang bermakna pada variasi 
volume pencucian terhadap pembacaan nilai absorbansi, namun secara deskriptif
adanya angka selisih pada masing-masing absorbansi akan sangat bermakna karena 
akan mempengaruhi nilai titer yang terkandung. Untuk mendapatkan hasil ELISA 
yang akurat disarankan untuk memilih komposisi buffer pencuci Tris-buffered 
saline (TBS) dan phosphate-buffered saline (PBS) yang mengandung 
0,05%(v/v)Tween 20 serta mengikuti saran teknik pencucian sesuai dalam kit 
ELISA yang digunakan.

ABSTRACT
The Effect of Washing Volume Variations on the Reading of Anti-HBsAg 
Absorbance Value ELISA Method

Hepatitis B is a disease that causes inflammation of the liver and is usually 
caused by the Hepatitis B Virus (HBV). HBsAg is a hallmark of infection while 
Anti-HBsAg is an antibody to HBsAg that appears when a patient has been infected 
with HBV for a long time or has been vaccinated against Hepatitis B. This study 
aims to determine the effect of washing volume variations on the reading of the 
absorbance value of Anti-HBsAg using the ELISA reader method. This type of 
research is an experiment with Posttest Only with Control Design. Samples were 
taken by purposive sampling technique on health workers at the Puskesmas 
Martapura Timur who had been vaccinated against hepatitis B as many as 3 people, 
carried out Anti-HBsAg examination based on variations in washing volume using 
the ELISA reader method. Data analysis used the Repeated ANOVA test. The 
results showed that the variations in the washing volume carried out were 100 µl, 
150 µl, 200 µl, 250 µl, and 300 µl at the statistical value of 0.224 (p> 0.05) proving 
that there was no significant effect on the volume variation on the absorbance value 
reading, but descriptively the difference in the value of each absorbance will be 
very significant because it will affect the value of the titer contained. In order to 
obtain accurate ELISA results, it is recommended to choose a washing buffer 
composition of Tris-buffered saline (TBS) and phosphate-buffered saline (PBS) 
containing 0.05%(v/v) Tween 20 and follow the suggested application techniques 
in the ELISA kit used.
