Perbedaan Morfologi Sel Eritrosit dan Sel Leukosit Menggunakan Buffer 
Fosfat pH 6,8 dan Air Mineral Metode Wright Stain

Metode pewarnaan Sediaan Apusan Darah Tipis (SADT) yang dianjurkan 
oleh The Internasional Council For Standardization in Haematology (ICSH) 
salah satunya adalah Wright Stain. Larutan pengencer yang digunakan dalam 
pewarnaan Wright adalah buffer fosfat. Buffer sebagai larutan pengencer Wright
harus diperhatikan ketepatan Power of Hydrogen (pH) untuk hasil pewarnaan 
yang baik. Larutan buffer fosfat pH 6,8 mudah didapatkan, namun harganya 
cukup mahal, Sehingga air mineral dalam kemasan digunakan sebagai larutan 
alternatif yang bisa digunakan sebagai larutan pengencer yang lebih murah dan 
mudah untuk diperoleh. Bertujuan untuk mengetahui perbedaan morfologi sel 
eritrosit dan sel leukosit Metode Wright Stain menggunakan Buffer Fosfat pH 6,8 
dan Air Mineral Metode Wright Stain. Jenis penelitian observasional laboratorium 
dengan rancangan comparative study dan 6 jenis air mineral dalam kemasan untuk 
3 level darah kontrol yaitu high, normal, dan low. Dengan 5 kali pengulangan 
dalam satu jenis darah kontrol. Hasil penelitian morfologi sel eritrosit dan sel 
leukosit menggunakan buffer fosfat pH 6,8 pada darah kontrol high, normal dan 
low adalah baik (100%). Sedangkan hasil penelitian morfologi sel eritrosit dan sel 
leukosit menggunakan air mineral memiliki hasil pewarnaan yang baik. 
Berdasarkan uji Friedman dari ketiga jenis darah kontrol mendapatkan nilai 
signifikansi p>0,05 berarti tidak terdapat perbedaan bermakna antara penggunaan 
buffer fosfat pH 6,8 dan air mineral. Sehingga dapat disimpulkan air mineral 
dengan pH 6,8–7,2 dapat digunakan sebagai alternatif larutan pengencer 
pengganti buffer fosfat pH 6,8. Saran untuk peneliti selanjutnya agar melakukan 
penelitian pada pemeriksaan malaria dengan Metode Wright menggunakan air 
mineral dalam kemasan sebagai larutan pengencer pengganti buffer.

ABSTRACT
The Difference in the Morphology of Erythrocytes and Leukocytes Using 
Phosphate Buffers pH 6.8 and Mineral Water with Wright Stain Method

One of the staining methods recommended by The International Council 
for Standardization in Haematology (ICSH) is the Wright Stain. The diluent used 
in Wright's stain is phosphate buffer. Buffer as Wright's diluent solution must be 
considered for the accuracy of Power of Hydrogen (pH) for good staining results. 
Phosphate buffer solution pH 6.8 is easy to obtain, but the price is quite 
expensive, so bottled mineral water is used as an alternative solution that can be 
used as a diluent solution that is cheaper and easier to obtain. Aims to determine 
the differences in morphology of erythrocyte and leukocyte cells. Wright Stain 
method using Phosphate Buffer pH 6.8 and Mineral Water using Wright Stain 
Method. This type of research is an observational laboratory with a comparative 
study design and 6 types of bottled mineral water for 3 control blood levels, 
namely high, normal, and low. With 5 repetitions in one type of control blood. 
The results of the study of morphology of erythrocytes and leukocytes using 
phosphate buffer pH 6.8 in high, normal and low control blood were good 
(100%). While the results of the research on morphology of erythrocyte and 
leukocyte cells using mineral water have good staining results. Based on the 
Friedman test, the three types of control blood obtained a significance value of 
p>0.05, meaning that there was no significant difference between the use of 
phosphate buffer pH 6.8 and mineral water. So it can be concluded that mineral 
water with a pH of 6.8–7.2 can be used as an alternative diluent to replace 
phosphate buffer with a pH of 6.8. Suggestions for further researchers are to 
conduct research on malaria examination with the Wright Method using bottled 
mineral water as a buffer substitute diluent solution.
