Metode pewarnaan Sediaan Apusan Darah Tepi (SADT) yang dianjurkan 
oleh The Internasional Council For Standardization in Haematology (ICSH) salah 
satunya adalah Wright Stain yang digunakan untuk pengamatan sel dan komponen 
sehingga dapat melihat morfologi sel darah. Ketepatan Power of Hydrogen (pH) 
buffer sebagai larutan pengencer Wright Stain harus diperhatikan untuk hasil 
pewarnaan yang baik. Larutan buffer fosfat pH 6,8 mudah didapatkan, namun 
harganya cukup mahal, sehingga air mineral digunakan sebagai larutan alternatif 
yang bisa digunakan sebagai larutan pengencer yang lebih murah dan mudah untuk 
diperoleh. Penelitian ini bertujuan untuk mengetahui perbedaan hasil morfologi sel 
eritrosit dan sel leukosit Metode Wright Stain menggunakan Buffer Fosfat pH 6,8 
dan Air Mineral Bermerk Lokal. Jenis penelitian observasional laboratorium 
dengan rancangan comparative study. Menggunakan 6 jenis air mineral bermerk 
lokal yang diproduksi wilayah Kalimantan Selatan dan 3 level darah kontrol yaitu 
high, normal, dan low. Pengulangan yang dilakukan 5 kali dalam satu jenis darah 
kontrol. Hasil penelitian morfologi sel eritrosit dan sel leukosit pada darah kontrol 
high, normal dan low menggunakan buffer fosfat pH 6,8 dan air mineral bermerk 
lokal A, B, C, D, E, dan F memiliki hasil pewarnaan yang baik. Berdasarkan uji 
Friedman dari ketiga jenis darah kontrol mendapatkan nilai signifikansi >0,05 
berarti tidak terdapat perbedaan bermakna antara penggunaan buffer fosfat pH 6,8 
dan air mineral bermerk lokal A, B, C, D, E, dan F. Sehingga dapat disimpulkan air 
mineral bermerk lokal A, B, C, D, E, dan F dapat digunakan sebagai larutan 
pengencer alternatif pengganti buffer fosfat pH 6,8.

ABSTRACT
The Differences in Morphology of Erythrocyte and Leukocyte Cells Wright 
Stain Method Using Phosphate Buffer pH 6.8 and Local Branded Mineral 
Water

One of the staining methods for Peripheral Blood Smear (SADT) 
recommended by The International Council for Standardization in Haematology 
(ICSH) is the Wright Stain which is used for observing cells and components so 
that they can see the morphology of blood cells. The accuracy of the Power of 
Hydrogen (pH) buffer as a diluent for Wright Stain must be considered for good 
staining results. Phosphate buffer solution pH 6.8 is easy to obtain, but the price is 
quite expensive, so mineral water is used as an alternative solution that can be used 
as a diluent solution that is cheaper and easier to obtain. Aims to determine 
differences in morphological results of erythrocyte and leukocyte cells. Wright 
Stain method using Phosphate Buffer pH 6.8 and Local Branded Mineral Water. 
This type of research is an observational laboratory with a comparative study 
design. 6 types of local branded mineral water were used, produced in the South 
Kalimantan region and 3 control blood levels, namely high, normal, and low. 
Repetition was carried out 5 times in one type of control blood. The results of the 
study of morphology of erythrocytes and leukocytes in high, normal and low control 
blood using phosphate buffer pH 6.8 and local branded mineral water A, B, C, D, 
E, and F had good staining results. Based on the Friedman test, the three control 
blood types obtained a significance value > 0.05 meaning there was no significant 
difference between the use of phosphate buffer pH 6.8 and local branded mineral 
water A, B, C, D, E, and F. So it can be concluded that water local branded minerals 
A, B, C, D, E, and F can be used as an alternative diluent to replace phosphate buffer 
pH 6.8.
